He was the eldest son of carpenter František FAMFULÍK and Helena FAMFULÍKOVÁ, née ŠIMKOVÁ. He trained as a miller, then served with the 8th Legionary Regiment in Místek and after military service helped his parents on the farm in Vidlatá Seč. After that he pursued a career in the gendarmerie.

During the Protectorate, he was ordered to arrest the communist functionary Josef Formánek. Instead, he warned him and provided him with a gun and grenades. Formánek was later arrested and during interrogation he revealed Famfulík as his informant.
Famfulík was arrested, interrogated and sentenced to death in Berlin. He was imprisoned in Terezín and Prague together with three other gendarmes.
On July 8, 1943, he was publicly executed in Kobylisy, Prague. The execution was intended to intimidate the Protectorate gendarmes. Before the execution, the convicts were stripped of their rank insignia and all four refused to be blindfolded. Their bravery surprised the German officers. The execution was watched by 150 gendarmes and police officers.

František Famfulík was married to Maria TOPERCEROVÁ (1915-1973) and they had a son, Vladislav (1940-1981).
